Bengkoang (Pachyrhizus erosus) has been traditionally used as sun
screening and skin whitening. However, the active compounds in
bengkoang which have skin whitening and sun screening activity have
not been discovered yet.Most important for maintaining skin whiteness
is to avoid ul traviolet exposure by using sunscreen [Sayre and
Nadkarni, 1994], because UV radiation can induce formation of reactive
oxygen species (ROS) [Matsuura et al., 2006] and hydroxyl radical
[Wang et al., 2006]. These radicals induce tyrosinase and enhance
melanin biosynthesis [Matsuura et al., 2006]. Furthermore, the radical
scavengers as antioxidant may reduce hyperpigmentation and can be
also used as whitening materials [Seo, et., 2003; Karg et a., 2006]. The
other important way to reduce melanin biosynthesis is by inhibi ting
tyrosinase [Khatib et al., 2005]. The combination of sunscreen,
antioxidant and anti tyrosinase compounds in skin whitening cosmetics
is needed to get a synergic affect. Structure elucidation and biological
activity test of the active compounds of bengkoang extract have been
carried out. A new compound (8,9-furanyl-pterocarpan-ol) has been
selected as a biomarker for extract s tandardization. The optimum of
HPLC condition for standardization is column (Zorbax SB-C18; i.d. 0.46
cm; 5 ?m particle size), eluent (gradient elution using MeOH-water),
detector (UV-detector at 293 nm). The obtained LOD value is 0.51 ±
0.02 µg. The potentials of this compound to absorb UV r ay, antioxidant
and anti tyrosinase are 4.018 mAU*S/mml ; 2.113±0.001mM (SC50);
7.19±0.11 mM (IC50), respectively.
