Objective: This study was aimed to develop sensitive, selective, simple methods for determine the levels of, catechin , caffeine, and epigallocatechin gallate in green tea leaves extract. Method: The analytical separation was used on C-18 column (X-Bride®, Waters, 250 x 4.6µm), column temperature was set on 15 oC, The mobile phase used ortho-phosphoric acid 0.1%: water: acetonitrile: methanol (4:76:10:10 v/v/v/v) pH 3.6 and UV detector at a wavelength of 274 nm. Green tea extract was extracted by water extraction and fractioned using ethyl acetate. Results: The retention time of epigallocatechin gallate, catechin, and caffeine were 6.994, 5.310, and 5.956 minutes, respectively and calibaration curves were linear with a coeffecient correlation (r) of 0.999 for epigallocatechin gallate, 0.998 for catechin, and 0.999 for caffeine. The selectivity, precision, accuracy, and recovery were validated in green tea extract.
