Curcumin analogue PGV-1 shows cytotoxic effect and induces apoptosis in WiDr colon cancer cells. PGV-1 inhibitis G2M phase and increases the degree of polyploidy in T47D breast cancer cell. Moreover, it has been revealed that the inhibition of this G2M phase progression correlated with tubulin polymerization. Thea aim of this research was to determine whether PGV-1 induces G2M arrest and polyploidy in WiDr cells. The cells were treated with PGV-1 12.5, 25, 50, and 100 uM for 24, 48, and 72 h, respectively. The cells distribution was determined by FACS Beckton Dickinson and the data was analyzed by using Modfit Lt software. The results showed thta PGV-1 performed G2M phase inhibition in concentration dependent manners. Moreover, PGV-1 increased the level of cyclin A and cyclin B, indicating inhibition of G2M phase progression. However, PGV-1 did not induce polyploidy in WiDr cells. |
