Eria hyacinthoides orchid is a natural orchid from Indonesia that has distinctive scented flowers but the 14 characterisation of phytochemical content that has the potential to be used as biomedical materials and 15 bio fragrance in this type of orchid is not yet known. This study aims to determine the morphology, 16 improve in vitro growth through somatic embryogenesis and analyse the phytochemical content of E. 17 hyacinthoides orchid leaves. The methods include planting leaf explants on Knudson C (KC) media with 18 the addition of ZPT variations of NAA (1 mg/L) and TDZ (1 mg/L), and extraction of phytochemicals 19 from leaves by multistage maceration with qualitative tests of secondary metabolites. The results showed 20 that somatic embryos were formed on leaf explants after 3 months of cultivation and the presence of 21 alkaloids, flavonoids, and tannins in E. hyacinthoides leaf extract was detected
