Incorporation of Ovalbumin into Carbonate Apatite as a Candidate for Protein Delivery

Introduction. Appropriate biomaterial and controlled size particle are the important component to achieve effective delivery system. Reducing size of the particle is recommended because it can overcome the barriers during cellular uptake. Biomimetic carbonate apatite (CHA) is now considered as candidate for protein delivery because it has high affinity to protein, high biocompatibility and biodegradibility, and increases protein stability. In this study, nano-CHA was prepared and ovalbumin (OVA) protein was incorporated into the CHA particles. Experimental. Biomimetic CHA granules (sized 150-250µm) were synthesized by simple wet precipitation method in our laboratory. The effect of stirring speed, temperature, and maturation time in reducing size of CHA granules were investigated. There were no surfactant nor dispersing agents added to the solution. The effect of CHA and OVA protein concentrations on particle size of CHA-OVA complexes were also investigated. Particle size distribution was determined using particle size analyzer (PSA). The morphology of CHA-OVA complexes was examined with transmission electron microscopy. The CHA-OVA complexes were characterized using Fourier transform infrared. Result and Discussion. The range of distribution in particle size of CHA was narrower in the experiment with the lower stirrer speed, higher temperature, and elongation maturation in solution. After the incorporation of protein, particle size of CHA-OVA complexes were influenced by the concentration of CHA and OVA protein. Particle size of CHA-OVA complexes significantly increased because the addition of protein. It was as expected because protein tends to aggregate around the isoelectric point. The addition of the protein resulted in micro-sized particle of CHAOVA complexes. TEM photograph of CHA-OVA complex showed that particles are spherical and uniform. Characterization of CHA-OVA complex using FTIR showed that calcium ions from CHA bind into COO- groups of protein. Conclusions. The present study described the simple method for incorporating protein ovalbumin into CHA particles and proposed that this can be subsequently used to incorporate protein for effective delivery system. The ex-vivo and in-vivo delivery systems are under investigation in our laboratory.
