Abstrak/Abstract Background: Aedes aegypti is a primer vector of dengue virus (DENV) and chikungunya virus (CHIKV). The mosquito susceptibility toward DENV and CHIKV depended on recognitions receptor pathogen. The C-type lectins (CTLs) were known as an important role as mediator virus infection. Thus, the objective of this study is to determine potential receptor and binding affinity of protein-protein interaction between CTLs and viruses envelope (DENV-1, 2, 3, 4, and CHIKV), based on in silico analysis. Materials and Methods: The sample sequences were available from GenBank (NCBI), ten CTLs depended on vectorbase accession no. Homology modeling based on a minimum standard at 20% that processed in the SWISS-MODEL server. Simulation and affinity interaction energy processed in the ClasPro server, which used the protein-protein docking method. The visualization interaction among molecules presented in form tertiary protein structure, that was processed in PyMol V1.7.4. Results: The homology modeling shown all of 3D protein model scores there are over 20%. The higher homology modeling score is available by CHIKV envelope protein (KF925315.1) at 98.72%. The candidate receptor of CTL based on lowest interaction affinity energy, CTL GenBank ID: AAEL014382-PA, is the lowest score at -1407.20 kcal/mol. Conclusion: Based on homology modeling and lowest interaction energy could be referred for study of the wet laboratory. This information is expected, giving an illustration that CTLs receptor facilitated DENV and CHIKV co-infection at Aedes aegypti
