Identification of Proteolytic Lactic Acid Bacteria Isolated from Colostrum of Dairy Cattle by 16S rRNA Sequence Analyses

Abstrak/Abstract
Aim: Lactic acid bacteria (LAB) produces lactic acid, bacteriocins, and peptide compounds with beneficial biological properties. We aimed to identify LAB isolated from dairy cow colostrum with superior proteolytic properties.
Methods: The isolates were biochemically identified by 16S rRNA sequence analyses. The procedures include the isolation of the bacteria from the colostrum, followed by qualitative and quantitative selection, to obtain the optimal protease enzyme activity, and then a low pH resistance analysis.
Results: Lactic acid bacteria isolation, selection, and identification were analyzed via descriptive and quantitative analyses using one-way ANOVA for Completely Randomized Design. We isolated 6 superior proteolytic LAB isolates based on the protease enzyme activities, as follows: BAL Pro 4, BAL Pro 3, BAL Pro 8, BAL Pro 9, BAL Pro 21, and BAL Pro 20. The selected isolates were then identified molecularly by using genes for 16S rRNA sequence analysis.
Conclusion: The isolates identified molecularly showed similarities with Lactobacillus fermentum in BAL Pro 8, 9, and 20 and Limosilactobacillus fermentum in BAL Pro 3 and 21.
