IDENTIFICATION FLAVONOID BY THIN LAYER CHROMATOGRAPHY AND CALCULATION TOTAL FLAVONOID, TOTAL FENOLIC OF TAGETES ERECTA L LEAVES BY SPECTROPHOTOMETRY

Tagetes erecta L commonly known as bunga tahi kotok and empirically
been widely used as a tradi tional medicine. Ethanol extract of T erecta
as raw material medicine should be standardized by applying one of the
womb as a marker compound. Qualitatively identification of flavonoid
and fenolic performed by Thin Layer Chromatography (TLC).
Determination of total flavonoid levels equivalent quercetin and total
fenolic equivalent gallic acid ware conducted using spectrophotometry.
The results of the identification flavonoid in TLC using mobile phase
Chloroform: ethyl acetate: methanol: glacial acetic acid (7: 2: 0.5: 0.5)
with the spray reagent AlCl3, samples obtained Rf value 0.33 and Rf
value of quercetin as a comparison was 0.35. Identification of fenolic
using mobile phase chloroform : acetic acid (7 : 3), samples obtained Rf
value 0.25 and Rf value of gallic acid as a comparison was 0.29. The
result of the calculation total flavonoid equivalent quercetin performed
by spectrophotometric method with a regression equation y =
0,00194x - 0.00754 and correlation coefficient r2 = 0.99901 results
obtained 1.38% w/w. The result of the calculation total fenolic
equivalent gallic acid with a regression equation y = 0,00485x + 0.0000
and correlation coefficient r2 = 0.99755 results obtained 2,9% w/w.
