Domperidone, an antiemetic drug, has a low oral bioavailability. Nausea-vomiting conditions, especially in pediatrics, need a convenient and effective administration manner. A transdermal patch is a dosage form attached to the skin non-invasively, circumventing the first-pass effect and giving practical administration. Optimization of the developed pharmaceutical formulations is an important step to obtaining the patch's desired physical properties.
The purposes of this study were: 1) to formulate domperidone into patch dosage form with the variation of oleic acid (OA) and isopropyl alcohol (IPA) as an enhancer, 2) to understand the effect of various concentrations of enhancer on the physical properties, and 3) to determine the optimum formula composition of the patch.
The optimization of the patch was performed based on the Simplex-Lattice-Design method using Design-Expert version 10 software. Evaluations of the patch's physical characteristics were based on parameters of weight variation, patch thickness, folding endurance, moisture content (MC), moisture uptake (MU), and content uniformity.
The results indicate that OA and IPA concentrations' variation influenced weight variation, patch thickness, and content uniformity. Meanwhile, such compositions do not influence MC, MU, and folding endurance. OA has a dominant effect on weight, thickness, and MU. Meanwhile, IPA has a dominant impact on MC and content uniformity. The optimum formula was 1% OA and 25% IPA in the formula of the patch.
