Abstrak/Abstract Chemotherapy as the most cancer treatment that has been used nowadays has many harmful side effects including suppressed the immune system (immunosuppressive). A method that can be done to overcome this problem is giving immunostimulator agent as co-chemotherapy. According to in vitro and in vivo researches, polysaccharides contained in Ling Zhi (Ganoderma lucidum) had been confirmed having many immunostimulatory activities. The aim of this research was to determine the immunostimulatory activity of water extract of G.lucidum by observe the relative CD8+ T lymphocyte cell percentage on Sprague Dawley female rats induced by doxorubicin. Extraction was carried out by infusion method. Sprague Dawley female rats were divided into 6 treatment groups, they were (A) 2,33 ml/kgBW doxorubicin as control, (B) ā€œIā€ commercial product + doxorubicin as comparing control, (C) 100 mg/kgBW G.lucidum extract + doxorubicin, (D) 450 mg/kgBW G.lucidum extract + doxorubicin, (E) 450 mg/kgBW G.lucidum extract as extract control, and (F) without treatment control. Treatment was done for seven days. The extracts and ā€œIā€ commercial product were given per oral daily, while doxorubicin were injected intra peritoneal on 1st and 4th day. Blood samples were taken before treatment (0th day) and on 7th day. Relative CD8+ T lymphocyte cell percentages were obtained by flowcytometry. The data were analyzed statistically using paired sample t-test and one way ANOVA continued by Post Hoc test. The result showed that the water extract of G.lucidum dose 100 mg/kgBW and 450 mg/kgBW increased the relative CD8+ T lymphocyte cell percentage on rats induced by doxorubicin. The water extract of G.lucidum has potency to be developed as co-chemotherapy immunostimulatory agent.
