Abstrak/Abstract BACKGROUND: The use of the herb Andrographis paniculata (Burm.F). Ness and Centella asiatica L. Urban herb are reported can improve the insulin resistance in type-2 diabetes mellitus. However, the molecular mechanisms of those combined plants in lowering blood glucose are unknown. This present study was aimed to examine effects of A. paniculata herb purified extracts (APHPE) and C. asiatica herb purified extracts (CAHPE) combined on PPARγ and GLUT4 expression at mRNA level in cell culture system using 3T3-L1 adipocyte in vitro. In insulin signalling, GLUT4 acts as a transporter on glucose uptake outside the cells. METHODS: 3T3-L1 adipocyte cells were used to investigate gene expression detected using the reverse transcription polymerase chain reaction method. The adipocyte cells were differentiated by using insulin, dexamethasone and IBMX from 3T3-L1 cells. Pioglitazone, APHPE, CAHPE and the purified extracts combination were tested on glucose uptake activity, PPARγ and GLUT4 expression. RESULTS: The results showed that combination of APHPE 30 μg/ml and CAHPE 10 μg/ml can increase glucose uptake characterized by increased PPARγ and GLUT4 expression at mRNA levels significantly relative to compared to negative control (DMSO), and has not significant difference compared to pioglitazone 0.02 μM and each purified single extract. CONCLUSION: The combination of APHPE and CAHPE can increase glucose uptake and insulin sensitivity through up-regulation of PPARγ and GLUT4 expression at mRNA levels.
