Abstrak/Abstract Abstract: Nuclear factor kappa B (NF-kB) p105/50 has been shown play an important role in pathological cancer. Ov erexpression of NF-kB p50 contribute to cell migration and metastasis of breast cancer. Triple-negative breast cancer (TNBC) is highly aggressive with poor prognosis. Moreover, TNBC is often resistant to cytotoxic chemotherapy. The characteristic of TNBC is activation of NF-kB signaling. Therefore, inhibition of NF-kB signaling can block metastas is of TNBC. The aim of this study is evaluating the effect of curcumin on NF-kB p105 RNA expression in TNBC cell line and in silico study to explicate the possible mechanism of action curcumin against NF-kB precursor protein p105. 4T1 cells were cultured and isolated. Expression of NF-kB by reverse transcription-polymerase chain reaction (RT-P CR). The X-ray crystallographic structure of the NF-?B precursor p105/50 was extracted from protein data bank (PD B). 2-D structures of curcumin were retrieved from PubChem. Molecular docking simulation study was carried out by using molecular operating environment (MOE) 2015. The NF-kB p105 RNA expression in 4T1 breast cancer cell wa s significantly decreased by curcumin treatment. In silico study show, the benzene in curcumin structure can interact with Ile142 by hydrogen binding with value of docking score -13,7709. To sum up that curcumin effectively suppress NF-kB p105/50 gene expression, a marker of metastatic, through interaction with amino acid of NF-kB. Further rese arch is going on to prove its potential as a therapeutic agent for TNBC.
