Abstrak/Abstract SMN protein is one type of protein that must be present in every human. If SMN1 genes do not function, the production of SMN protein is insufficient. This condition triggers Spinal Muscular Atrophy (SMA) disease. Several therapies that focus on increasing the SMN protein production in patients with SMA disease have been developed. In this study, the SMN Protein biosensor has been successfully extended to determine the therapy’s effectiveness given to patients with SMA disease. Quartz Crystal Microbalance (QCM) active layer coated with SMN Antibody for enhanced sensitivity for SMN protein detection. This experiment uses a flow system because the sample is blood plasma (concentration in percent %). QCM-D biosensor coated with additional SMN Antibody has good sensitivity. The sensitivity was 21.1 Hz/%, higher than the undoped one (13,8 Hz/%) for SMN protein detection in the blood sample. The frequency shift of plasma samples from healthy subjects is higher than that of patients with SMA disease. Therefore, this result was also supported by dissipation monitoring that showed interactions between SMN protein in the blood plasma and different functionalized surfaces on the QCM electrode.
