Detection and Analysis of Fruit Ripening Coding Genes of Strawberry Plants (Fragaria x ananassa) Treated Indole-3-acetic acid Induction

Strawberry are among Indonesia’s most popular commodities with a high economic value. The quality of strawberries is indicated by its ripeness. The hormone auxin serves to encode such fruit ripening genes as FaPYR1 and FaCHS. The purpose of the present study was to detect FaPYR1 and FaCHS genes in strawberries induced by indole-3-acetic acid (IAA). The methods were planting strawberries (Fragaria x ananassa ‘Crystal’ and ‘KP Brite’), induction by IAA in three different concentrations (10 ppm, 30 ppm and 60 ppm), genomic DNA isolation, quantitative testing, FaPYR1 and FaCHS amplification with PCR and morphological data analysis using SPSS version 16. Results showed that the concentration of 30 ppm produced the highest results in terms of the leaf length, leaf width, stem height, and stem diameter. The highest number of leaves was found in concentration of 60 ppm. The lowest morphological result was found in the control. The highest concentration of genomic DNA was found in the red-stage fruits treated with 30 ppm and the lowest concentration was found in the white stage at 60 ppm. The PCR amplification products showed positive results for FaCHS in all treatments and negative results for FaPYR1 in several treatments
