Abstrak/Abstract | Reactive oxygen species (ROS) exposure could lead to aging of the human cell. Tomato (Solanum lycopersicum L.) and carrot (Daucus carota L.) callus ethanolic extract are flavonoid-rich. Meanwhile tomato and carrot callus aqueous extract is suggested to contain protein that plays a role in cell regeneration against free radical. This study was aimed to evaluate the cytoprotective activity of tomato and carrot callus in preventing hydrogen peroxide (H2O2)-induced cell cycle arrest in G0/G1 phase and inhibiting cell death. Cell death analysis of Human Dermal Fibroblast adult (HDFa) cells was performed using double staining method. Cell death inhibition showed in 0.15 mg · mL?1 and 0.25 mg · mL?1 of tomato callus ethanolic extract (TCEE); 0.15 mg · mL?1 and 1.0 mg · mL?1 of tomato callus aqueous extract (TCAE); 0.15 mg · mL?1 and 0.25 mg · mL?1 of carrot callus ethanolic extract (CCEE); 0.05 mg · mL?1 and 0.50 mg · mL?1 of carrot callus aqueous extract (CCAE) pre-treatment compared with H2O2 control. The result of cell cycle analysis using FACS flows cytometry showed that 1.0 mg · mL?1 TCAE, 0.15 mg · mL?1 CCEE and 0.5 mg · mL?1 pre-treatment inclines to reduce accumulation of cell in G0/G1. |
