Strawberry (Fragaria spp.) is the result of domesticated plants originating from Chile, America. Indonesian local strawberries have less quality than those developed abroad, Indonesian strawberries have relatively small fruits and taste very sour. The efforts to find an improved quality local strawberry have been made by the Indonesian Citrus and Subtropical Fruits Research Institute (ICSFRI), but there are no many researches to characterize and identify the ploidy level of the local strawberry plants conducted in Indonesia. The objective of this research was to investigate the strawberry chromosome character of strawberry cultivar collections that are cultured in ICSFRI as the effort to improve local strawberry gene. This study began from November 2014 to October 2015. Strawberry root sampling was carried out in ICSFRI located in the Tlekung Village, Junrejo Sub-district, Batu, East Java. Chromosome preparation was made of the root tips with squash method using acetoorcein 1%. Flow cytometry analysis was conducted at Animal Physiology Laboratory of Brawijaya University using a flow cytometer. The results of the research showed that strawberry cultivars studied can be classified into two groups according to chromosome number (ploidy), they are diploid and tetraploid.Californica and Earlibrite cultivars are the groups of diploid (2n = 2x = 14) and Aerut, Berastagi, Festival, Holland and Rosa Linda belong to the group of tetraploid (2n = 4x = 28).The absolute length of chromosome strawberry of various cultivars is ± 0.9-2.9 µm. Shapes of strawberry chromosome are dominated by metacentric and submetacentric chromosomes. |
