Abstrak/Abstract Introduction: Study on the discovery of traditional medicine has been done for years to solve people health problem. There are so many potential natural resources from Indonesia which could be observed for drug discovery. Objectives: This present study aimed to (i) quantify the total phenolic and flavonoid contents, (ii) examine the DPPH radical scavenging and alpha-glucosidase inhibitory activities, and (iii) investigate the correlation of the total phenolic and flavonoid contents of different parts of durian plants (Durio zibethinus Murr.) with their activities. Methods: The root, twig, inner fruit bark and leave of durian plants (Durio zibethinus Murr.) were macerated with 70% of ethanol. The total phenolic content was measured using a Folin-Ciocalteu assay and the total flavonoid content using colorimetric aluminum chloride assay. The DPPH free radical scavenging and alpha-glucosidase inhibitory activity were performed in vitro using microplate reader. Additionally, we also compared their activities with their total phenolic and flavonoid contents which were determined quantitatively using spectrophotometer UV-Vis. Results: The root, twig, inner fruit bark and leave extracts revealed a difference DPPH free radical scavenging and alpha-glucosidase inhibitory activities. The root and twig extracts exhibited the strongest DPPH free radical scavenging activities among samples evaluated with IC50 values of 18.54±0,58 and 96.65±0,14 µg/mL; and alpha-glucosidase inhibitory activity with IC50 values of 0,58±0,01 and 1.55±0,02 µg/mL. The root and twig extract also contained the highest phenolic compounds accounting of 95.63±0.032 and 31.65±0,132 Gallic acid Equivalent (GAE)/g dry extract. The total phenolic content of the four extracts has a significant correlation with their DPPH free radical scavenging and alpha-glucosidase inhibitory activity. Conclusion: In this study, we found that the root and twig extract of Durio zibethinus Murr. were potential source to develop as the new antioxidant and alpha-glucosidase inhibitor in the treatment of diabetes.
