Diabetes mellitus (DM) is a metabolic disorder with symptoms of chronic
hyperglycemia and disturbances in metabolism of carbohydrates, fats and
proteins. Due to insulin deficiency, insulin resistance or inability of ? cells of
pancreas to produce insulin.. This study was aimed to evaluate the effects of
ethanolic extract from Sargassum polycystum on glucose level and lipid profile
in streptozotocin-induced diabetic rats. S. polycystum from Binangeun
Seashore, Banten was extracted with ethanol 96% then tested for total phenolic
content of extract, and analysied using TLC and GC-MS. Ethanolic extract of S.
polycystum at 150 mg/kg, and 450 mg/kg was administered orally to
streptozotocin induced diabetic rats, once daily for 15 days. The results
obtained were then compared with normal controls (non-diabetic + normal
saline 0,9% p.o), negative control (diabetic + CMC-Na 0,5% p.o) and positive
control (diabetic + glibenclamide 5 mg/kg p.o). The glucose levels, lipis profile
and body weight were measured on normal condition (baseline), on 0, 5th,
10th, 15th day, and pancreatic histopathological study was done on 15th day.
Total phenolic content of S. polycystum extract was 0,3 g GAE/100 g extract
and the extract contains terpenoids and phenolic such as camphor, elemol,
beta-elemene, alpha-curcumene, germacrone and phenol which might be
potential as oral antidiabetis therapy. S. polycystum extract with 150 mg/kg
concentrations had the highest capability on prevention of Diabetic Mellitus
indicated by the increase in body weight, decreased of blood glucose values,
increased of cholesterol values, decreased of triglyceride values, increased of
HDL-c and LDL-c. Necrosis was found in all streptozotocin-induced rats. S.
polycystum extract with 450 mg/kg concentrations showed highest reduction of
? cells of the pancreas induced by streptozotocin is better than S. polycystum
extract with 150 mg/kg concentration. |
