Abstrak/Abstract Introduction: Ultraviolet radiation induces skin photoaging by decreasing collagen consequently altering TGF-/Smad pathway. Achatina fulica mucous (AFM) is a natural remedy containing anti-aging agents. Objective: To investigate effects of A. fulica on UVB-induced fibroblast photoaging by assessing TGF-, Smad3 and Smad7 mRNA expressions. Methods: The mucous was extracted from 50 Achatina fulica snails taat were stimulated by a 5-10Volt electricity shock for 30-60 seconds and converted into powder by the freeze-drying process. The human dermal fibroblast cell culture was divided into six groups: group 1 were normal fibroblasts without UVB irradiation as normal control, groups 2-5 consisted of 100mJ/cm2 UVB induced aged fibroblasts. Group 2 had no treatment as negative control, group 3 was treated by PRP 10% as positive control group and groups 4-6 were treated by various concentrations of Achatina fulica mucous (3.9µl; 15.625 µl and 62.5 µl). The TGF-, Smad3 and Smad7 mRNA expressions in the different NHDFs groups were assessed by quantitative polymerase chain reaction. Results: The mRNA expressions of TGF- and Smad3 increased meanwhile Smad7 decreased significantly in the Achatina fulica groups contrast with UVB group. Concerning the concentration of Achatina fulica mucous, the 62.5 µL gave the best result. Conclusion: Achatina fulica mucous reduces Smad7 however increases Smad3 and TGF-beta mRNA expressions.
